Der ELISA Kit für menschliches DEFa1 (Defensin Alpha 1, Neutrophil) ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von DEFa1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes DEFa1 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes DEFa1 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die humanes DEFa1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration des humanen DEFa1 in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
