Das Human F12 (Gerinnungsfaktor XII) Mikrosample ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von F12.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes F12 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes F12 ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes F12, einen Biotin-konjugierten Antikörper und ein Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem F12 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
