Das Human PLA2G2D(Phospholipase A2, Gruppe IID) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von PLA2G2D.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes PLA2G2D ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für humanes PLA2G2D ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes PLA2G2D, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem PLA2G2D in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
