Das Humane VIM (Vimentin) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von VIM für Forschungszwecke.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes VIM ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für humanes VIM ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes VIM, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem VIM in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
