Das Maus ADAM8(A Disintegrin And Metalloprotease 8) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von ADAM8.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus ADAM8 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus ADAM8 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Maus ADAM8, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus ADAM8 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
