Das Human F2 (Gerinnungsfaktor II) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von F2 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Human F2 ist. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte zugegeben, anschließend ein biotin-konjugierter Antikörper, der spezifisch für Human F2 ist. Danach wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Human F2, den biotin-konjugierten Antikörper und das enzymkonjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human F2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
