Das Ratten-NOS1(Nitric Oxide Synthase 1, Neuronal) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von NOS1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Ratten-NOS1-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-NOS1 ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Ratten-NOS1, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-NOS1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
