Das Humane DPP4 (Dipeptidyl-Peptidase IV) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von DPP4.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Human-DPP4 ist. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Human-DPP4 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Human-DPP4, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human-DPP4 in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
