Das Ratten-GIP(Gastric Inhibitory Polypeptide) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von GIP.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte ist mit Ratten-GIP vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein biotin-konjugierter Antikörper, der spezifisch für Ratten-GIP ist, hinzugefügt. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung wird die Enzym-Substrat-Reaktion durch Zugabe von Schwefelsäurelösung gestoppt und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-GIP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
