Das Maus-AngII (Angiotensin II) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von AngII.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper gegen Maus-AngII vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-AngII ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nachdem die TMB-Substratlösung hinzugefügt wurde, zeigen nur die Vertiefungen, die Maus-AngII, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-AngII in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
