Das Human-CCL1(Chemokine C-C-Motif Ligand 1)-ELISA-Kit ist ein ELK-Biotechnology-ELISA-Kit zur Forschungsmessung von CCL1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für humanes CCL1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein biotin-konjugierter Antikörper, der spezifisch für humanes CCL1 ist. Als Nächstes wird in jede Mikrotiterplattenvertiefung Avidin, konjugiert an Meerrettichperoxidase (HRP), gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes CCL1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem CCL1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
