Das Human S100A10 (S100 Calcium Binding Protein A10) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von S100A10.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes S100A10 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für humanes S100A10 ist. Danach wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes S100A10, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem S100A10 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
