Das Humane HSPA8 (Heat Shock 70kDa Protein 8) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von HSPA8.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes HSPA8 ist. Standards oder Proben werden dann zusammen mit einem biotin-konjugierten Antikörper, der spezifisch für humanes HSPA8 ist, zu den entsprechenden Mikrotiterplattenvertiefungen gegeben. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes HSPA8, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem HSPA8 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
