Das Maus-ACTH(Adrenocorticotropic Hormone) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von ACTH zu Forschungszwecken.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-ACTH ist. Standards oder Proben werden dann zusammen mit einem biotin-konjugierten Antikörper, der spezifisch für Maus-ACTH ist, in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die Maus-ACTH, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-ACTH in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
