Das Human D2D (D-Dimer) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von D2D zu Forschungszwecken.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes D2D ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für humanes D2D ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die humanes D2D, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem D2D in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
