Das Maus-IFN-Gamma (Interferon Gamma) Mikrosampel-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsmessung von IFN-Gamma.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-IFN-Gamma ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann ein biotin-konjugierter Antikörper, der spezifisch für Maus-IFN-Gamma ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus-IFN-Gamma, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe einer Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-IFN-Gamma in den Proben wird dann durch Vergleich der optischen Dichte (OD) der Proben mit der Standardkurve bestimmt.
