Das Maus PRAP1 (Proline-rich acidic protein 1) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von PRAP1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem spezifischen Antikörper gegen Maus PRAP1 vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein Biotin-konjugierter Antikörper, der spezifisch für Maus PRAP1 ist, hinzugefügt. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus PRAP1, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus PRAP1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
