Der Hund TNFa (Tumornekrosefaktor Alpha) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von TNFa.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Hund TNFa ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Hund TNFa ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Hund TNFa, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Hund TNFa in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
