Das Maus-SPA(Surfactant Associated Protein A) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungs-Messung von SPA.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus-SPA ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplatten-Vertiefungen gegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus-SPA ist. Als Nächstes wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplatten-Vertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Maus-SPA, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-SPA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
