Das Humane PCSK6 (Proprotein Convertase Subtilisin/Kexin Typ 6) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von PCSK6 in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für Human PCSK6 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, danach ein Biotin-konjugierter Antikörper, der spezifisch für Human PCSK6 ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Human PCSK6, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human PCSK6 in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
