Das Pferde-SAA (Serum Amyloid A) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Erforschung und Messung von Pferde-SAA.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Pferde-SAA ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Pferde-SAA ist. Als Nächstes wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Pferde-SAA, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Pferde-SAA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
