Das Schweine-CLU(Clusterin)-ELISA-Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsanalyse von CLU.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Schweine-CLU vorbeschichtet. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen zugegeben, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für Schweine-CLU ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung zugegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Schweine-CLU, einen biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Schweine-CLU in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: Clusterin functions as an extracellular chaperone that helps prevent aggregation of non-native and stress-damaged proteins.
