Das Ratten-ANXA1-(Annexin A1)-ELISA-Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ANXA1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-ANXA1 ist. Standards oder Proben werden dann zusammen mit einem Biotin-konjugierten Antikörper, der spezifisch für Ratten-ANXA1 ist, zu den entsprechenden Mikrotiterplattenvertiefungen gegeben. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Ratten-ANXA1, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-ANXA1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Annexin A1 (ANXA1) is a calcium- and phospholipid-binding annexin family protein with established roles in inflammatory control, innate and adaptive immune responses, membrane organisation and cytoskeletal signalling.
Biological function: ANXA1 binds calcium and phospholipid membranes and acts as an anti-inflammatory regulator. Reviewed UniProt evidence supports roles in glucocorticoid-mediated control of inflammation, T-cell signalling and differentiation, phagocytosis, actin-cytoskeleton organisation and regulation of exocytosis.
