Das Hunde-HMGB-1 (High Mobility Group Protein B1) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von HMGB-1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für Hunde-HMGB-1 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Hunde-HMGB-1 ist. Anschließend wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Hunde-HMGB-1, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung gestoppt und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Hunde-HMGB-1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
