Das Pig TIMP1 (Tissue Inhibitors Of Metalloproteinase 1) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von TIMP1.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper beschichtet, der spezifisch für Schweine-TIMP1 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Schweine-TIMP1 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbveränderung, die Schweine-TIMP1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Schweine-TIMP1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
