Der Humane POSTN (Periostin) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von POSTN in der Forschung.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes POSTN ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend wird ein biotinylierter Antikörper, der spezifisch für humanes POSTN ist, hinzugefügt. Danach wird in jede Vertiefung der Mikrotiterplatte mit Meerrettichperoxidase (HRP) konjugiertes Avidin gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbveränderung, die humanes POSTN, den biotinylierten Antikörper und das enzymkonjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem POSTN in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
