Das Humane IGFBP5 (Insulin Like Growth Factor Binding Protein 5) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsbestimmung von IGFBP5.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für Human-IGFBP5 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Human-IGFBP5 ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Human-IGFBP5, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human-IGFBP5 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
