Das Maus-PAI2 (Plasminogen-Aktivator-Inhibitor 2) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Forschungsmessung von PAI2.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Maus-PAI2 vorbeschichtet. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen zugesetzt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Maus-PAI2 ist. Anschließend wird Avidin, konjugiert mit Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Maus-PAI2, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-PAI2 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
