Der Human-CTSL(Cathepsin L)-ELISA-Kit ist ein ELK-Biotechnologie-ELISA-Kit zur Forschungsmessung von CTSL.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes CTSL ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für humanes CTSL ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes CTSL, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem CTSL in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
