Der Human-TP73(Tumorprotein p73)-ELISA-Kit ist ein ELK-Biotechnologie-ELISA-Kit zur Forschungszwecken dienenden Messung von TP73.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem für humanes TP73 spezifischen Antikörper vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem biotin-konjugierten Antikörper, der für humanes TP73 spezifisch ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die humanes TP73, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem TP73 in den Proben wird dann durch Vergleich der optischen Dichte (OD) der Proben mit der Standardkurve bestimmt.
