Der Human LIAS (Lipoyl Synthase) ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von LIAS.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humane LIAS ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für humane LIAS ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen eine Farbänderung, die humane LIAS, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration der humanen LIAS in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
