Das Rat ESM1 (Endothelial Cell Specific Molecule 1) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ESM1.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-ESM1 ist. Standards oder Proben werden zu den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-ESM1 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur die Vertiefungen, die Ratten-ESM1, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-ESM1 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
