Das PROA (Protein A) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von PROA.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte ist mit PROA vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für PROA ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von PROA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
