Der Human Anti-RyR (Anti-Ryanodin-Rezeptor-Kalziumfreisetzungskanal-Antikörper) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Erforschung und Messung von Anti-RyR.
Dieser Assay verwendet die kompetitive Inhibition mittels Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit Human Anti-RyR vorbeschichtet. Standards oder Proben werden den entsprechenden Vertiefungen der Mikrotiterplatte hinzugefügt, gefolgt von einem Biotin-konjugierten Antikörper, der spezifisch für Human Anti-RyR ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung gestoppt, und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human Anti-RyR in den Proben wird anschließend durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
