Das Maus OGA (O-GlcNAcase) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungsmessung von OGA.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus OGA ist. Standards oder Proben werden den entsprechenden Mikrotiterplattenvertiefungen zugegeben, dann mit einem Biotin-konjugierten Antikörper, der spezifisch für Maus OGA ist. Als Nächstes wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus OGA, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten, eine Farbveränderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus OGA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
