Der Human MYO15A (unkonventionelles Myosin-XV) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Messung von MYO15A in der Forschung.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte ist mit einem Antikörper vorbeschichtet, der spezifisch für humanes MYO15A ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, anschließend ein Biotin-konjugierter Antikörper, der spezifisch für humanes MYO15A ist. Danach wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die humanes MYO15A, biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MYO15A in den Proben wird dann durch Vergleich der OD-Werte der Proben mit der Standardkurve bestimmt.
