Product description

Background

Survivin is a member of the Inhibitor of Apoptosis (IAP) gene family and contains a single BIR domain. Increased survivin expression at the G2/M checkpoint contributes to tissue homeostasis and differentiation during embryonic and fetal development. Increased expression in differentiated adult tissues, however, frequently results in unchecked cell survival, resulting in cancer. Survivin can protect cells from caspase-induced apoptosis activated in mitosis presumably by binding to Caspase-3, -7, and -9.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human Survivin antibody. Survivin present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human Survivin Antibody is added and binds to Survivin in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated Survivin antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human Survivin. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

BIRC5baculoviral IAP repeat containing 5

View target guide

BIRC5 (baculoviral IAP repeat containing 5), widely known as survivin, is an inhibitor-of-apoptosis-family protein studied for its roles in mitotic progression, chromosome segregation and cell-survival regulation.

Biological function: BIRC5 encodes survivin, a single-BIR-domain member of the inhibitor of apoptosis protein family. Beyond regulation of apoptotic cell death, survivin is a component of mitotic chromosome-passenger machinery and contributes to accurate chromosome segregation and cytokinesis. Multiple transcript variants encode distinct isoforms.
