Product description

Background

Carbonic anhydrase 1 is an enzyme that in humans is encoded by the CA1 gene. Carbonic anhydrases (CAs) are a large family of zinc metalloenzymes that catalyze the reversible hydration of carbon dioxide. They participate in a variety of biological processes, including cellular respiration, calcification, acid-base balance, bone resorption, and the formation of aqueous humor, cerebrospinal fluid, saliva, and gastric acid.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Bovine CA-1 antibody. CA-1 present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Bovine CA-1 Antibody is added and binds to CA-1 in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CA-1 antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Bovine CA-1. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

CA1carbonic anhydrase 1

View target guide

Carbonic anhydrase 1 (CA1; carbonic anhydrase I/CA-I, also CAB) is a cytosolic zinc metalloenzyme that catalyses reversible carbon dioxide hydration. It is especially abundant in erythrocytes and is studied in CO2 transport, bicarbonate chemistry and systemic acid-base physiology.

Biological function: CA1 catalyses the reversible interconversion of carbon dioxide and water with bicarbonate and protons through a zinc-containing active site. This rapid reaction supports carbon dioxide handling and proton/bicarbonate balance; its high erythrocyte abundance makes CA1 useful for studying carbonic-anhydrase catalysis and red-cell gas and acid-base chemistry.
