Product description

Background

N epsilon carboxymethyl lysine (CML or Carboxymethyl Lysine) is formed by the non enzymatic Schiff base reaction of glucose with proteins, followed by an Amadori rearrangement and oxidation that leaves only a carboxymethyl group attached to the lysine. The levels of CML adducts accumulate over time and have been used as an indicator of both serum glucose levels and oxidative protein damage. Elevated serum CML modified proteins have been associated with diabetes and may contribute to diabetic retinopathy, nephropathy and angiopathy.

Test principle

This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Human CML antibody. CML present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Human CML Antibody is added and binds to CML in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated CML antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Human CML. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Research target context

BCRBCR activator of RhoGEF and GTPase

View target guide

BCR encodes breakpoint cluster region protein, a multidomain regulator of small GTPase signalling. This ABMIUM target record connects the human BCR reference identifiers with catalogue products that target BCR.

Biological function: BCR combines opposing regulatory activities toward small GTP-binding proteins. Its C-terminal GAP domain promotes GTP hydrolysis by RAC1, RAC2 and CDC42, while its central DH domain has guanine-nucleotide exchange activity toward CDC42, RHOA and RAC1. The amino terminus also has intrinsic kinase activity. BCR is additionally important because chromosomal rearrangement with ABL1 produces the BCR-ABL fusion associated with chronic myeloid leukaemia and other leukaemias.
