12-HETE (12-Hydroxyeicosatetraensäure) ELISA Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsbestimmung von 12-HETE.
Dieser Assay verwendet die kompetitive Enzymimmunoassay-Technik. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit 12-HETE vorbeschichtet. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann mit einem biotin-konjugierten Antikörper, der spezifisch für 12-HETE ist. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Danach wird TMB-Substratlösung hinzugefügt. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von 12-HETE in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
