Das Ratten-CD45(Cluster of Differentiation 45) Microsample ELISA Kit ist ein ELK Biotechnology ELISA Kit für die Forschungsmessung von CD45.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-CD45 ist. Standards oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-CD45 ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen, die Ratten-CD45, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-CD45 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
