Das Ratten-ATG3 (Autophagy Related Protein 3) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ATG3.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Ratten-ATG3 ist. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Ratten-ATG3 ist. Anschließend wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbveränderung, die Ratten-ATG3, biotin-konjugierten Antikörper und enzymkonjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbveränderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-ATG3 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.

Biological function: ATG3 is an E2 conjugating enzyme in the autophagy machinery. It catalyses covalent conjugation of ATG8-family proteins, including LC3 and GABARAP family members, to phosphatidylethanolamine-containing membrane lipids. ATG3 cycles between ATG7-mediated loading and the ATG12-ATG5-ATG16L1 E3-like complex to promote ATG8 lipidation and formation of autophagic isolation membranes.
