Das Maus-BPA(Bisphenol A) ELISA Kit ist ein ELK Biotechnology ELISA Kit zur Forschungs-Messung von BPA.
Das in diesem Kit angewandte Testprinzip ist ein Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem spezifischen Antikörper gegen Maus-BPA vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, gefolgt von einem biotin-konjugierten Antikörper, der spezifisch für Maus-BPA ist. Anschließend wird Avidin, das mit Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Maus-BPA, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus-BPA in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
