Das Maus ISG15(Interferon Stimulated Gene 15) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von ISG15.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für Maus ISG15 ist. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, dann mit einem biotin-konjugierten Antikörper, der spezifisch für Maus ISG15 ist. Als Nächstes wird mit Meerrettichperoxidase (HRP) konjugiertes Avidin zu jeder Mikrotiterplattenvertiefung gegeben und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus ISG15, biotin-konjugierten Antikörper und enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet, und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus ISG15 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
