Das Ratten-KRAS(V-Ki-Ras2 Kirsten Rat Sarcoma Viral Oncogene Homolog) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von KRAS.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem für Ratten-KRAS spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Vertiefungen der Mikrotiterplatte gegeben, danach ein Biotin-konjugierter Antikörper, der für Ratten-KRAS spezifisch ist. Anschließend wird Avidin, das an Meerrettichperoxidase (HRP) konjugiert ist, zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur jene Vertiefungen eine Farbänderung, die Ratten-KRAS, den Biotin-konjugierten Antikörper und das Enzym-konjugierte Avidin enthalten. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Ratten-KRAS in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
