EasyStep Human NGAL (Neutrophil Gelatinase Associated Lipocalin) ELISA Kit ist ein ELK Biotechnology ELISA-Kit zur Forschungsbestimmung von EasyStep Human NGAL.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper beschichtet, der spezifisch für humanes NGAL ist, und die Wells der humanen NGAL-Standardplatte, die mittels proteinbezogener Techniken vorbeschichtet wurden, werden separat bereitgestellt. Standard-/Probenverdünnungspuffer oder Proben werden zu den entsprechenden Mikrotiterplatten-Wells hinzugefügt, dann wird ein HRP-konjugierter Antikörper, der spezifisch für humanes NGAL ist, hinzugefügt. Nach Zugabe der TMB-Substratlösung zeigen nur jene Wells, die humanes NGAL und einen HRP-konjugierten Antikörper enthalten, einen Farbwechsel. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem NGAL in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
