Das EasyStep Human MIG(Monokine Induced By Interferon Gamma) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von EasyStep Human MIG.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper beschichtet, der spezifisch für humanes MIG ist, und die Wells der Standardplatte für humanes MIG, die mit proteinbezogenen Techniken vorbeschichtet wurden, werden separat bereitgestellt. Standard-/Probenverdünnungspuffer oder Proben werden in die entsprechenden Wells der Mikrotiterplatte gegeben, dann wird ein HRP-konjugierter Antikörper, der spezifisch für humanes MIG ist, hinzugefügt. Nach Zugabe der TMB-Substratlösung zeigen nur die Wells, die humanes MIG und den HRP-konjugierten Antikörper enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und der Farbwechsel spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem MIG in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
