Das EasyStep Human VASP(Vasodilator Stimulated Phosphoprotein) ELISA Kit ist ein ELISA Kit von ELK Biotechnology zur Forschungsmessung von EasyStep Human VASP.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper vorbeschichtet, der spezifisch für humanes VASP ist, und die humanen VASP-Standardplattenvertiefungen, die mittels proteinbezogener Techniken vorbeschichtet wurden, werden separat bereitgestellt. Standard-/Probenverdünnungspuffer oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann wird ein HRP-konjugierter Antikörper, der spezifisch für humanes VASP ist, hinzugefügt. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die humanes VASP und den HRP-konjugierten Antikörper enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von humanem VASP in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
