Das Maus KRT14 (Keratin 14) Mikrosample ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von KRT14.
Das in diesem Kit angewendete Testprinzip ist der Sandwich-Enzymimmunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Maus-KRT14-spezifischen Antikörper vorbeschichtet. Standards oder Proben werden in die entsprechenden Mikrotiterplattenvertiefungen gegeben, dann mit einem Biotin-konjugierten Maus-KRT14-spezifischen Antikörper. Als Nächstes wird Avidin, konjugiert an Meerrettichperoxidase (HRP), zu jeder Mikrotiterplattenvertiefung hinzugefügt und inkubiert. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Maus KRT14, Biotin-konjugierten Antikörper und Enzym-konjugiertes Avidin enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Maus KRT14 in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
