Das EasyStep Human MPO (Myeloperoxidase) ELISA Kit ist ein ELISA-Kit von ELK Biotechnology zur Forschungsmessung von EasyStep Human MPO.
Das in diesem Kit angewandte Testprinzip ist der Sandwich-Enzym-Immunoassay. Die in diesem Kit enthaltene Mikrotiterplatte wurde mit einem Antikörper beschichtet, der spezifisch für Human MPO ist, und die Human MPO Standardplattenvertiefungen, die mittels proteinbezogener Techniken vorbeschichtet wurden, werden separat bereitgestellt. Standard/Probenverdünnungspuffer oder Proben werden zu den entsprechenden Mikrotiterplattenvertiefungen hinzugefügt, dann wird ein HRP-konjugierter Antikörper, der spezifisch für Human MPO ist, hinzugefügt. Nach Zugabe der TMB-Substratlösung zeigen nur diejenigen Vertiefungen, die Human MPO und den HRP-konjugierten Antikörper enthalten, eine Farbänderung. Die Enzym-Substrat-Reaktion wird durch die Zugabe von Schwefelsäurelösung beendet und die Farbänderung wird spektrophotometrisch bei einer Wellenlänge von 450 nm ± 10 nm gemessen. Die Konzentration von Human MPO in den Proben wird dann durch Vergleich der OD der Proben mit der Standardkurve bestimmt.
